how I open the port 25_ I have problem with SMTP, I have CISCO router 1801 and ASA 5510?

Pleas help me, I have problem with the port 25, I have cisco1801 router and cisco ASA 5510 and sophos. I do not see where is the problem if into the ISP, ASA or sophos?
