Need help with my iTunes account in resetting file folder

I have accidently removed the progam file that directs my music to my iTunes account by moving it to an external hard drive to free up space on my PC hard drive. When I realized my mistake, I put the file back onto my PC hard drive but iTunes doesn't recognize or find that folder now, please help!
